ELISA Tests for Fungi
Application:
The test can be used to detect the Fungi in manyhost plant tissues infected with Phytophthora spp., such as leaves, stems, roots,seeds, and potato tubers, as well as in pure fungal cultures.
Reactivity:
This ELISA detects at least the following 16 Phytophthoraspecies: P. cactorum, P. cambivora, P. capsici, P. cinnamomi, P. citricola,P. citrophthora, P. cryptogea, P. drechsleri, P. erythroseptica, P. fragariae,P. infestans, P. megasperma, P. palmivora, P. parasitica, and P. sojae. Itreacted with all 39 isolates of Phytophthorainfestans tested.
Sensitivity:
Sensitivity of the ELISA is moderately high. The fungus can be consistently detected in infected plant tissues diluted at1:270 - 1:810.

A rabbit polyclonal antibody for capture and amonoclonal antibody for detection.
Specificity:
This test system reacts negatively with thefollowing species of Fungi: Pythium aphanidermatum, P. catenulatum, P.heterothalicum, P. irregulare, P. oligandrum, P. spinosum, P. splendens, P.torulosum, P. ultimum, P. ultimum var. ultimum. The test didnot react with isolates of Aspergillussp., Fusarium sp., Moniliniasp., Penicilliumsp., Rhizoctonia sp., and Sclerotiniasp. There are moderate cross-reaction occurring with pure culture extracts of Pythiumaristosporum, P. arrhenomanes, P. graminicola, P. paroecandrum, and P. violae,and weak cross-reaction occurring with pure culture extracts of Pythiummultisporum, P. myriotylum, P. sylvaticum, P. tardicrescens, P. vanterpoolii.
