The Live/Dead Fixable Staining Kits have been engineered for differentiating between living and deceased cells by utilizing flow cytometry or fluorescence microscopy. Live/dead stainings serve as valuable tools in the examination of protein expression on cell surfaces through flow cytometry as it helps to exclude the intracellular fluorescence signals emanating from dead cells with permeable plasma membranes.
Live/Dead Fixable Stains function as amine-reactive dyes that cannot penetrate cell membranes. These dyes possess the capability to seep into dead cells which have compromised membranes, thereby labeling free amines on proteins located within the cell. For living cells, the dyes latch on to proteins on the surface, but these are considerably less numerous than proteins within the cell, resulting in a lesser degree of staining compared to dead cells. The dye labeling is markedly stable which allows for cell fixation and permeabilisation without the loss of fluorescence or dye transfer between cells. The protocol for Live/Dead Fixable Staining has been optimized to enhance live/dead differentiation while minimally staining live cells. To avoid disrupting immunostaining, we provide a range of eight varied Live/Dead Fixable Stains for optimal versatility in multi-color analysis.
