Description:

Nitrofurans (furazolidone, furaltadone, nitrofurazone, and nitrofirantion) are synthetic chemotherapeutic agents with a broad antimicrobial spectrum against both gram-positive and gram-negative bacteria. In 1991, FDA withdrew several approved food animal nitrofuran products because of concerns regarding their carcinogenicity. AMOZ (5-methyl-morpholino-3-amino-2-oxazolidinone) is one of the key metabolites of nitrofurans and is a marker residue for furaltadone adulteration. Nitrofurans are also on the banned drug list in Canada.

Competitive Enzyme immunoassay

Detection Principle:

This test kit is based on the solid phase enzyme-linked immunosorbent assay (ELISA) for the detection of AMOZ. An unknown amount of AMOZ present in the sample and the fixed amount of AMOZ antigens pre-coated on the wells of microtiter plate/strips compete for the anti-AMOZ antibodies, which in turn is detected with enzyme conjugate. After incubation the wells are washed and the bound enzyme is visualised by adding TMB solution. Any coloured product is measured at 450 nm after adding stop solution. The absorbance value of the developed colour is inversely proportional to the amount of the AMOZ in the sample. The quantity of AMOZ in the test sample can be interpolated from the standard curve constructed from the standards, and corrected for sample dilution.

Introduction:

Furaltadone (AMOZ) is one of the nitrofurans, which are synthetic broad-spectrum antibiotics. Nitrofurans have been banned on treating animals used for food production, because of the possibility of increased cancer risk if people are exposed to them over a long period of time.
