Aflatoxin M1 is a metabolite of aflatoxin B1 and can be found in milk and milk related products, minor mycotoxin of Aspergillus flavus are also present in the milk of sheep and cow fed toxic meal. It is a member of the difurocoumarocyclopentenone series family, polycyclic aromatic compounds consisting of a cyclopenten-2-one ring that is fused to the difurocoumarin skeleton via a coumarin moiety. The following aflatoxin M1 ELISA kit is reliable and accurate protocol which is frequently being used for many applications. Aflatoxin M1 have been widely linked to liver damage and cancer which is the result of activating P450 to the epoxide causing the DNA to alkylate. The eventual toxicity is dependent on the balance of the liver enzymes that either activate it (Cytochrome P450) or those that detoxify it (Glutathione-S-Transferase). Aflatoxins are essentially naturally occurring mycotoxins which are be produced by a number of different species for example Aspergillus (in particular Aspergillus flavus and Aspergillus parasiticus).

Detection Principle:

This test kit is based on the solid phase enzyme-linked immunosorbent assay (ELISA) for the detection of Aflatoxin M1. An unknown amount of Aflatoxin M1 present in the sample and the fixed amount of Aflatoxin M1 antigens pre-coated on the wells of microtiter plate/strips compete for the anti-Aflatoxin M1 antibodies, which in turn is detected with enzyme conjugate. After incubation the wells are washed and the bound enzyme is visualised by adding TMB solution. Any coloured product is measured at 450 nm after adding stop solution. The absorbance value of the developed colour is inversely proportional to the amount of the Aflatoxin M1 in the sample. The quantity of Aflatoxin M1 in the test sample can be interpolated from the standard curve constructed from the standards, and corrected for sample dilution.

Intended Use:

Aflatoxin M1 ELISA kit is a quantitative procedure for analysing aflatoxin M1 levels present in milk or milk products. This assay has a minimum analytical sensitivity limit of < 10.0 pg/ml.

Introduction:

Aflatoxin M1 is the principal hydroxylated aflatoxin metabolite present in the milk of dairy cows fed a diet contaminated with Aflatoxin B1. Aflatoxins are carcinogenic and are highly implicated in hepatocellular carcinoma in humans.
